Detection of GSK-3β and NF-κB protein expression, after 48 h of treatment with GSK-3β inhibitors, by western blot assay. ALL cells were cultured in the presence of LiCl (10 mM) or SB216763 (10 μM) for 48 h. Cytosolic and nuclear fractions were prepared from the indicated samples. β-Actin and histone were used as markers for the purity of the cytosolic and nuclear fractions, respectively. GSK-3β inhibition led to depletion of GSK-3β nuclear pool in ALL cells, whereas nuclear levels of NF-κB p65 remained unchanged.
