The recombinant virus TPV/Δ 66R/Δ 2L/fliC in culture. The plaque morphology associated with the TPV as visualized using the fluorescent reporters EGFP and mCherry present in the recombinant ablated for both 66R and 2L, the TPV/Δ66R/Δ2L/fliC. (a) A phase contrast view of the plaque in an OMK cell monolayer at days 2, 4 and 6 post-infection, visualized in white light. (b) The same plaque viewed at excitation and emission wavelengths which allow EGFP to be visualized. (c) The same plaque viewed at excitation and emission wavelengths which allow mCherry to be visualized. Microscope used was a Nikon Diaphot, total magnification 400X, exposed for 1.5 seconds. Image capture was performed with QImaging color camera using MetaMorph software (version 7.6.0.0, Molecular Devices). Scale bar = 100 μm.
