T-type Ca2+ channel antagonists, mibefradil and NNC-55-0396 modulated phospho-p44/42 MAP kinase activation in MOLT-4 cells. a MOLT-4 cells were incubated with different concentration of mibefradil or NNC-55-0396 for 48 h. b-c MOLT-4 cells were treated with 10 μΜ Mibefradil or NNC-55-0396 for various time-points from 0 to 24 h. Cell lysates were separated by SDS-PAGE and transferred to PVDF membranes. Membranes were probed with a phosphospecific Ab to detect activated Erk1/2 (top panel). The membrane was stripped and reprobed with an Ab directed against Erk1/2 to detect the total amount of kinase loaded in each lane (middle panel). GAPDH was used as a loading control for each lane (bottom panel). The results are presented as mean ± SEM of three independent experiments. *P < 0.05, **P < 0.01 compared with untreated control cells
