From: PCBP1 depletion promotes tumorigenesis through attenuation of p27Kip1 mRNA stability and translation

Fig. 3

Overexpression of PCBP1 represses cell cycle progression and promotes apoptosis. (a) Flow cytometry analysis of cell cycle progression in A2780 and DLD-1 cells stably transfected with PCBP1 or PCBP1 with additional p27 knockdown. (b) The effect of PCBP1 and p27 in cell cycle progression of DLD-1 and A2780 cells. The populations of cells in G1, S and G2 phases are presented based on three independent experiments. *P <0.05, **P <0.01, ***P <0.001, ns indicates no difference (c) Flow cytometry analysis of cell cycle progression in A2780 and DLD-1 cells stably transfected with PCBP1 and the control cells treated with 0.08 μM MTX plus serum-free starvation for 24 h, and the whole medium was added for another 12 and 16 hours, respectively, for cell cycle progression analysis. (d) Representative immunoblot of the cleaved caspase-3 in PCBP1 overexpressing A2780 and DLD-1 cells. Cells were treated with 3 μM Paclitaxel for 24 hours in advance of immunoblotting analyses. (e) Flow cytometric analyses of apoptotic cells in A2780 and DLD-1 cells with PCBP1 overexpression, compared with their control cells. Cells were treated with 3μM Paclitaxel for the indicated time. Total percentage of Annexin V-positive cells is shown. Results are representative data from three independent experiments
