RER1 enhances metastasis and stem-cell like behavior of PC cells. (a and b) Knockdown of RER1 significantly reduced migration of Mia PaCa-2 cells and overexpression of RER1 significantly increased migration of PANC-1 cells (scratch wound assay). (c) Transwell assays were performed to determine the migration and invasion of RER1 overexpression or knockdown Mia PaCa-2 cells. (d) Transwell assays were performed to determine the migration and invasion of RER1 overexpression or knockdown PANC-1 cells. (e) H&E staining of the metastatic nodules in the lung of RER1 overexpression or knockdown PANC-1 cells. Tail vein injection of PANC-1 cells into nude mice (100X scale bars) was performed and the incidence of lung metastasis in the mice was counted. (f) Tumorsphere formation of RER1 overexpression or knockdown PANC-1 cells was performed. The total tumorsphere numbers in each well were counted and images were taken at 40× magnification. (g) Tumor incidence of RER1 overexpression or knockdown PANC-1 cells was counted. Cells were injected into the flank of mice with limiting dilutions as indicated. The number of tumors formed in each group was counted after 4 weeks. (h) CD133 positive cell population in RER1 overexpression or knockdown PANC-1 cells was determined by flow cytometry.
