Extracellular vesicles (EV) secreted by normal-cultured (N-EV) or hypoxia pre-challenged mesenchymal stem cells (MSCs) promote macrophage M2 polarization in vitro. Human monocytes freshly isolated from peripheral blood were induced with 20 ng/ml of M-CSF for 5 days with or without the presence of MSC-EV. a–d, flow cytometry determining the percentage of CD163+CD206+ cells or CD40+CD86+ cells among total CD68+ cells after induction. e–h, ELISA detecting protein expression of IL-10, TGF-β, CCL-18 and VEGF-α in macrophage culture medium after induction. Macrophages induced without MSC-EV were used as negative control (NC). Statistical analysis results by Student’s t test was marked by # and that by Dunnett’s test were marked by *. * or #, p < 0.05; **, p < 0.01; ***, p < 0.001
