FBXO22 promotes HCC cell growth by down-regulating the levels of p21 and affects cell cycle and apoptosis induced by DNA damage in vitro. (a and b) FBXO22-depleted HLF and HepG2 cells were transfected with the indicated constructs, total protein was extracted and subjected to western blotting using anti-FBXO22, anti-p21, or anti-GAPDH antibodies. Knockdown of p21 expression effectively promoted cell growth (c and d) and foci formation (e and f). The results are expressed as the means ± standard error of the mean (SEM) of three independent experiments. (e) HLF cells infected with the indicated lentiviral shRNAs were transfected with the indicated constructs for 24 h. Cells were stained with propidium iodide and analyzed using flow cytometry. The error bars represent the mean ± SD of three independent experiments. (f) HLF cells were infected with the indicated lentiviral shRNAs. Cells were then treated with either 0.2 μM doxorubicin (Dox) for 48 h, followed by flow cytometry analysis. The error bars indicate the mean ± SD of three in- dependent experiments
