TRPM7 silencing inhibits the EMT in ovarian cancer cells. (a) The relative levels of EMT molecule expression in SKOV3-sh, SKOV3-TRPM7-sh, OVCAR3-sh and OVCAR3-TRPM7-sh cells were determined by Western blot and qRT-PCR. (b) The levels of F-actin expression in those cells were determined by fluorescent microscopy after staining with Phalloidin-iFluor 488. (c) Immunofluorescent analysis of E-cadherin and Vimentin expression in ovarian cancer cells after stained with rabbit anti-E-cadherin and mouse anti-Vimentin and subsequent Alexa Fluor™488-conjugated goat anti-mouse IgG and Alexa Fluor™ 594-conjugated goat anti-rabbit IgG as well as DAPI. (d) The cellular morphology of SKOV3-sh, SKOV3-TRPM7-sh, OVCAR3-sh and OVCAR3-TRPM7-sh cells. (e) Western blot analysis of the relative levels of EGF-induced E-cadherin, N-cadherin, Vimentin, Twist expression in those cells. (f) The levels of E-cadherin, N-cadherin, Vimentin, Twist expression in the lung metastatic ovarian cancer tissues were determined immunohistochemistry.
