MiR-139 suppresses HCC growth partially through targeting KPNA2. Hep3B and SMMC7721 cells were transfected with miR-139 mimics and a construct encoding KPNA2 or an empty vector. Cell viability of (a) Hep3B and (b) SMMC7721 cells were measured with MTT assay at indicated time points. Apoptosis of (c) Hep3B and (d) SMMC7721 cells was measured by annexin V/Hoechst 33258 double staining flow cytometry. c and (d) SMMC7721 cells were transfected with miR-139 mimic oligos and a construct encoding DNMT1 or an empty vector. c Cell viability and (d) apoptosis were measured by MTT assay and flow cytometry, respectively. e Colonogenic, (f) migratory and (g) invading capacities of cells were assayed as described above. In cells transfected with (h) miR-139 mimics or (i) KNPA2 specific siRNAs, total and nucleus c-myc and POU5F1 were measured with western blot. j Co-IP assay detected the direct interaction between KPNA2 and c-myc and POU5F1.
