CD80 upregulation is mediated by STAT3 activation. CT26 cells were transfected with control or STAT3 siRNAs. After 24 h, cells were treated with 200 μM H2O2 for 24 h before qReal Time PCR (a) or flow cytometry (b) for CD80. (c) Flow cytometry analysis of CT26 cells after 1 h of preincubation with 5 μM 5,15-DPP and treatment with H2O2 (200 μM) for 24 h. (d) CT26 cells were transfected with plasmid vector alone (CTRL) or to overexpress constitutively active STAT3 (STAT3C); 24 h after transfection, cells were treated with 200 μM H2O2 for additional 24 h before flow cytometry for CD80. Flow cytometry analysis of (e) HT-29 and (f) HCT 116 cells after 1 h of preincubation with 5 μM 5,15-DPP and treatment with H2O2 (200 μM) for 24 h.
