M802 activated T cells and induced the secretion of cytokines in vitro. (a and b) SK-BR-3 cells were incubated with PBMCs at an E:T ratio of 5:1 and with different concentrations of M802 or Herceptin or MCO101 for 48 h. The PBMCs were collected and stained with anti-CD3-FITC and anti-CD69-PE or anti-CD25-APC antibodies. The activation was determined by flow cytometry. The cell free supernatant was used to detect the cytokine release including IFN-γ (c), TNF-α (d), IL-2 (e) and IL-6 (f) by ELISA. (g) Secretion of cytokines by PBMCs induced by 10 μg/mL M802 and control antibodies. M802 significantly promoted the secretion of cytokines, especially IL-6 and IFN-γ
