TBM-induced autophagy is dependent on ROS/AMPK activation in CRC cells. a. SW480 and HCT116 cells were treated with indicated concentrations of TBM. The total and phosphorylation levels of AMPK were examined by immunoblotting. b. Immunoblot analysis of LC3, AMPK and p-AMPK expression in SW480 and HCT116 cells transfected with siNC or siAMPK for 24 h, followed by treatment with or without 10 μM TBM for another 24 h. c. SW480 and HCT116 cells were transfected with Vector or DN-AMPK for 24 h, followed by treatment with or without 10 μM TBM for another 24 h. Immunoblot analysis of LC3, AMPK and p-AMPK expression in CRC cells. d-e. SW480 and HCT116 cells were treated with vehicle, TBM (10 μM), NAC (5 mM), or in combination for 24 h. ROS was detected by flow cytometric analysis (D), and the levels of cleaved-PARP1, p-AMPK, AMPK, Beclin1, Atg5 and LC3 were examined by immunoblotting (E). F. SW480 and HCT116 cells were transfected with GFP-LC3 for 48 h, and then treated as (D-E). GFP-LC3 puncta was shown and quantitated by immunofluorescence analysis. Scale bar, 10 μm. ***, p < 0.001
