A2aR knockdown provides sustained cytokine secretion by MSLN-CAR T cells in the presence of an A2aR agonist. Mock, MSLN-CAR and A2aR.KD1 and KD3.MSLN-CAR T cells were co-incubated with Hela cells in a 1:1 ratio or incubated in media, in the absence or presence of 1 μM NECA. The supernatant was harvested after 24 h and IFN-γ (a), TNF-α (b) and IL-2 (c) concentrations were measured by ELISA. Data are shown as mean ± SD. The results are representative of three independent experiments. *P < 0.05 by one-way ANOVA with Tukeyʼs post-test for multiple comparisons.
