Sensitivity to cytotoxic agents and targeted agents in vitro and in vivo. A. MTS cell survival assays were used to examine the sensitivity of the three new cell lines to commonly used drugs including 5-fluorouracil (5-FU), docetaxel, erlotinib, and RAD001 by treating 48 h at the indicated dosage. Cell viability and IC50 were determined as described in Materials and Methods. B. The three cell lines were treated with mitomycin and a mTOR inhibitor sapanisertib, for 48 h at the indicated dosage and subjected to MTS cell survival assays. Cell viability and IC50 were determined as described in Materials and Methods. C. The three cell lines were treated with JQ1 at the indicated dosage for 48 h, and cell survival was detected using MTS assay in vitro. D. Diagram showed the treatment plan in GA0518 PDX in vivo. E.
