PYCR1 inhibition increases bortezomib-mediated cell death in vitro and ex vivo. A OPM-2 and RPMI-8226 were cultured for 48 h in a hypoxic environment and treated with bortezomib and pargyline. Apoptosis was measured using flow cytometry by staining for Annexin V and 7-AAD (n = 5). B CD138+ MM cells from several patient samples were cultured for 24 h and treated with bortezomib and pargyline. Viability was measured by CellTiter Glo assay (n = 4). All experiments were performed in hypoxic conditions. ctrl = control, bz = bortezomib, par = pargyline. Significance was determined by one-way ANOVA. * p ≤ 0.05, ** p ≤ 0.01, *** p ≤ 0.001
