CDH17 and DSC1 regulate p120-catenin isoform 1 expression. A, B The indicated cell lines were transfected with control, DSC1 or CDH17 targeting siRNAs. Cell extracts were analyzed by Western blot to detect the indicated proteins. The indicated protein expression levels were significantly increased (*, p < 0.05; **, p < 0.01) or decreased (, p < 0.01; , p < 0.001) after siRNA transfection. C The same cell lines were transfected with empty vectors (Mock) or vectors encoding for CDH17 RAD, and the cell extracts analyzed as before. D, E The indicated transfectants were subjected to subcellular fractionation and analyzed by Western blot. RhoGDI, EGFR and Lamin B were used as loading controls for cytosolic, membrane and nuclear fractions, respectively. The expression levels of p120-catenin isoform 1 in each subcellular fraction were significantly increased (*, p < 0.05; **, p < 0.01) or decreased (, p < 0.05) after DSC1 silencing or CDH17 RAD overexpression. Data are representative of three independent experiments
