Adaphostin (ADA) induces nuclear localization of Nrf2 protein. (A) NCI-H522 cells were incubated for 2 h, 4 h and 6 h with 1 μM of ADA; nuclear and cytosolic proteins were resolved by Western blot analysis as indicated in the Materials and Methods (NS, non-specific band was used for normalization). Data are representative of three independent experiments. (B) Confirmation of ADA-induced Nrf2 translocation into the nucleus. NCI-H522 cells were incubated for 4 h or 6 h with 1 μM ADA and were stained with Nrf2 and FITC-conjugated antibodies. Nuclei were counter-stained with the fluorescence dye DAPI.
