ROS-YAP-JNK pathway is involved in WZ35 induced mitochondrial dysfunction. a MDA-MB-231 cells were cultured with WZ35 or curcumin for 12 h, and cellular oxygen consumption rate (OCR) was measured in real time using the Seahorse XF96 Extracellular Flux Analyzer after basal OCR was measured at three time points, followed by sequential injection of oligomycin (1 μM), FCCP (0.5 μM) rotenone (1 μM), and antimycin A (1 μM). The overall OCR curves were plotted as the mean OCR ± SD of three replicates. b Basal respiration, maximal respiration and spare respiration were assessed, respectively. c, d MDA-MB-231 cells were treated with or without WZ35 and co-treated with NAC. e, f MDA-MB-231 cells were overexpressed YAP and treated with or without WZ35. g, h MDA-MB-231 cells were treated with or without WZ35 and co-treated with JNK inhibitor SP600125.
