SLC-0111 increases the effect of Cis-Pt on tumor growth in vivo. A Nude mice bearing FaDu xenografts, after tumor establishment (50 mm3), were treated with vehicle, SLC-0111 (100 mg/kg per os administration, via oral gavage) or Cis-Pt alone (3 mg/kg per i.p. injection), or the two drugs in combination, as shown in treatment schedule. Tumor growth was monitored by calipers overtime and tumor volume mean ± SD of each group is reported. Mice body-weight was measured at the indicated days and the weight mean ± SD is shown. (*p < 0.0001 relative to vehicle; #p < 0.05 combinatorial treatment vs Cis-Pt alone) (B) At end of treatments nude mice bearing FaDu xenografts were injected with 2 nmol of Annexin Vivo 750 and subjected to imaging studies using FRI at 2 and 24 h. At end of imaging study, tumors were harvested for ex vivo FRI analysis. Representative images as shown. C Lysates from recovered tumors were immunoblotted with antibodies anti pro-caspase-3/cleaved-caspase-3 and PARP/cleaved-PARP. Equal loading was confirmed by immunoblot with anti-Actin antibody.
